GHK-Cu Peptide (Glycyl-L-Histidyl-L-Lysine Copper)
GHK-Cu is a naturally occurring tripeptide copper complex formed by the amino acids glycine (Gly), L-histidine (His), and L-lysine (Lys) bound to a copper(II) ion (Cu²⁺). This small molecule has a molar mass of ~340.4 g/mol in its uncomplexed form (C₁₄H₂₄N₆O₄) and forms a stable square-planar copper chelate with high water solubility (~131 g/L).
GHK-Cu exhibits multifaceted biochemical activity at physiological concentrations (picomolar to nanomolar) by modulating gene expression, cellular signalling, and extracellular matrix (ECM) dynamics. It binds copper via the imidazole nitrogen of histidine and peptide backbone nitrogens, forming a bioactive complex that influences collagen, elastin, and glycosaminoglycan synthesis.
Key technical and functional features:
Peptide sequence: Gly-His-Lys (tri-amino acid)
Copper complex: 1:1 ratio of peptide to Cu²⁺
Molecular weight: ~340.38 g/mol (without Cu); complexed form ~ranges with Cu²⁺ bound
Solubility: High aqueous solubility
Stability: Stable at physiological pH (~6.5–7.5), can dissociate outside this range
Biological & functional actions:
Collagen and ECM support: Stimulates fibroblast production of collagen (types I, III, IV, VII), elastin, and glycosaminoglycans, aiding skin elasticity and firmness.
Gene modulation: Influences expression of thousands of human genes related to repair, inflammation, and antioxidant pathways.
Wound healing & regeneration: Enhances angiogenesis, immune cell recruitment, and tissue remodelling, with documented activity in animal models.
Antioxidant & anti-inflammatory effects: Acts as a copper chaperone activating enzymes like superoxide dismutase (SOD) and reducing pro-inflammatory signalling.
Sources & usage notes:
GHK-Cu is widely used in cosmetic and dermaceutical products (INCI: Copper tripeptide-1) for anti-aging, skin repair, and hair growth support, though clinical results depend on formulation and concentration.
